Description

Assay Genie's cell-based ELISA kit is designed for the sensitive and precise detection of phosphorylated TIF-IA at serine 649 in cell lysates. The assay uses a colorimetric detection method with measurement at 450 nm and is reactive with human samples. The reported dynamic range is greater than 5000 Cells, and the kit supports work in the research area of Epigenetics and Nuclear Signaling.

The indirect ELISA format is described as lysate-free, high throughput, and sensitive, allowing measurement of relative amounts of phosphorylated TIF-IA in cultured adherent cells. The kit can be used to screen the effects of various treatments, inhibitors, or activators on TIF-IA phosphorylation. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method can be used to determine cell density.

TIF-IA, also known as RRN3, is a key regulator of ribosome biogenesis and protein synthesis, playing a role in cell growth and proliferation. Phosphorylation at serine 649 is known to modulate its activity, making it a target of interest for studies of cell signaling pathways in conditions such as cancer, metabolic disorders, and neurodegenerative conditions. The protein has a reported molecular weight of 53,315 Da and is encoded by the RRN3 gene (Gene ID: 54700, UniProt ID: Q9NYV6).
