TGF alpha Colorimetric Cell-Based ELISA Kit

From Assay Genie, this Cell-Based ELISA Kit is designed to detect TGF alpha expression in cultured cells using a colorimetric method with absorbance read at 450 nm. The assay is reactive with human, mouse, and rat samples and is supplied in a 96-well microplate format. It is described as a convenient, lysate-free, high-throughput and sensitive assay that can measure the relative amounts of TGF alpha in cultured cells and screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on TGF alpha.

Qualitative determination is achieved by an indirect ELISA format in which TGF alpha is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colour-producing reaction upon substrate addition. Because the Cell-Based ELISA is qualitative, a monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values. Following the colorimetric measurement, the Crystal Violet whole-cell staining method may be used to determine cell density, so that absorbance values can be normalised to cell amounts and plating differences adjusted.

The dynamic range is > 5000 Cells, and the kit is stored at 4 °C for 6 Months. TGF-Alpha is a key signalling molecule involved in cell growth, proliferation and differentiation, and its dysregulation has been implicated in cancer, inflammatory disorders and tissue fibrosis. The associated gene TGFA (Gene ID: 7039, UniProt ID: P01135) encodes a ligand for the epidermal growth factor receptor, activating a signalling pathway for cell proliferation, differentiation and development.
