The TFE3 Transcription Factor Activity Assay from Assay Genie quantifies the DNA-binding activity of transcription factor E3, a member of the microphthalmia-associated (MiT) transcription factor family implicated in lysosomal biogenesis, autophagy, and metabolic regulation. Rather than measuring protein abundance, the kit captures functionally active TFE3 that recognizes its consensus oligonucleotide sequence immobilized on a microplate well, with bound complexes reported through a colorimetric enzymatic readout. Nuclear and cell lysates prepared from human, mouse, or rat samples are loaded directly, and the workflow supports studies of TF activation following nutrient stress, mTORC1 modulation, or in translocation-driven disease models such as alveolar soft part sarcoma and MiT family renal cell carcinoma.
