Description

A colorimetric cell-based ELISA kit from Assay Genie designed for the precise and sensitive detection of TF2A1 levels in cell lysates and cell culture samples, with the product literature describing TF2A1 as a key transcription factor (Transcription Factor AP-2 alpha) that regulates gene expression and plays roles in cell growth, differentiation and development (S1). The kit uses a 96-well microplate format and an indirect ELISA approach that provides qualitative determination, in which the target is captured by specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition, with absorbance read at 450 nm (S1). It is described as a convenient, lysate-free, high-throughput and sensitive assay for measuring relative TF2A1 protein expression in cultured cells, and can be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on TF2A1 (S1).

Reactivity is reported for Human, Mouse and Rat samples, with a stated dynamic range of > 5000 Cells (S1). The kit is intended for adherent cells, and a monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values (S1, C0). A Crystal Violet whole-cell staining step can be used to determine cell density and adjust for plating differences (S1). Storage is specified at 4°C for 6 months (S1). Database identifiers for the target include gene symbol GTF2A1, UniProt ID P52655, Gene ID 2957 and OMIM 600520, with a reported molecular weight of 37,152 Da and chromosomal location 14q31.1 (S1). The cited research areas include cancer, developmental biology and neurobiology, reflecting the protein's role as a general transcription factor and a component of the RNA polymerase II transcription machinery (S1).
