Telomerase Colorimetric Cell-Based ELISA Kit

An In-Cell ELISA Kit from Assay Genie designed for the detection of Telomerase protein expression in cultured cells, intended for use with adherent cell samples. The kit provides a lysate-free, high-throughput, and sensitive colorimetric assay that can measure relative amounts of Telomerase in cells and screen for the effects of various treatments, inhibitors such as siRNA or chemicals, and activators on Telomerase. The qualitative determination is achieved by an indirect ELISA format, in which Telomerase is captured by Telomerase-specific primary antibodies and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, enabling HRP to catalyze a colorimetric reaction upon substrate addition.

For normalization, the kit includes a monoclonal antibody specific for human GAPDH as an internal positive control, and the Crystal Violet whole-cell staining method may be used to determine cell density so absorbance values can be normalized to cell amounts. Detection is colorimetric, read at 450 nm, and the kit has a dynamic range of > 5000 Cells. The target, TERT (telomerase reverse transcriptase), is a ribonucleoprotein polymerase that maintains telomere ends by addition of the telomere repeat TTAGGG and is implicated in cellular senescence and oncogenesis. The product is reactive with human samples and is stored at 4°C with a stability of 6 Months.
