Competitive immunoassay for tetracycline-class antibiotic residue screening in food matrices. The Assay Genie kit uses pre-coated coupled antigen on a 96-well microplate, where analyte in the sample competes with coated antigen for binding sites of an anti-tetracycline antibody, generating an inverse signal that rises as tetracycline concentration falls. HRP conjugate and TMB substrate deliver a chromogenic readout readable on a standard microplate reader.

Calibration is provided across six liquid standards (0, 0.15, 0.45, 1.35, 4.05, and 12.5 ng/mL), yielding a working detection range of 0.15 to 12.5 ng/mL under the kit protocol. Reported method sensitivity is 0.15 ppb (ng/mL), with documented detection limits of 3 ppb in muscle (method 1) and eggs, 100 ppb in muscle (method 2) and liver, 200 ppb in feed, and 10 ppb in raw and finished milk. Cross-reactivity testing against related tetracycline analogues shows chlortetracycline at 154%, terramycin (oxytetracycline) at 46.4%, and deoxytetracycline at 8.9%, enabling laboratories to flag total tetracycline-class burden in submitted samples. Average sample recovery is 90%±30%.

Each kit ships with ELISA microtiter plate (96 wells), HRP conjugate (7 mL), antibody working solution (7 mL), substrate reagents A and B (6 mL each), stop solution (6 mL), 20× concentrated wash buffer (25 mL), 20× concentrated sample solution (50 mL), plate sealers, and protocol manual. Sample preparation requires trichloroacetic acid reagent and a homogenizer; required instrumentation includes a microplate reader, centrifuge, oscillators, balance (0.01 g sensibility), and micropipettes covering 20-200 µL single-channel, 100-1000 µL single-channel, and 30-300 µL multichannel. Incubations run at 25°C for 30 minutes followed by 20 minutes. Store refrigerated at 4°C and avoid freezing any kit components; return unused microwells to the foil bag with desiccant.

Intended for research use only in food safety surveillance, the Assay Genie assay supports residue testing workflows verifying compliance with maximum residue limits across dairy, poultry, and livestock supply chains.
