The TBX2 Transcription Factor Activity Assay from Assay Genie is a DNA-binding ELISA-type kit that quantifies activated TBX2 bound to its consensus T-box response element in nuclear and cell lysates. TBX2 is a T-box family transcription factor that predominantly acts as a transcriptional repressor, with documented roles in cell cycle regulation, senescence bypass, and oncogenesis in breast carcinoma and melanoma, as well as in cardiac and limb morphogenesis during embryonic development.

This microplate-based assay uses an immobilized double-stranded oligonucleotide containing the TBX2 recognition motif, and bound transcription factor is detected with a TBX2-specific antibody followed by an enzyme-based chromogenic readout. The protocol accepts nuclear extracts prepared from cultured human, mouse, and rat cells, requires 5–50 µL of sample per well, and supports up to 96 Assays per kit. Reagents should be stored at -20 °C with minimal freeze-thaw cycles, and the kit is intended for research use only.
