Description

A cell-based ELISA from Assay Genie designed for the quantification of TBX2 protein levels in cell lysates and culture supernatants. The kit is positioned for research into TBX2, a T-box family transcription factor involved in the transcriptional regulation of genes required for mesoderm differentiation, with described relevance to development, cancer and other conditions.

Detection uses an indirect ELISA format in which TBX2-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. The format is a 96-Well Microplate with colorimetric detection at 450 nm, a dynamic range of > 5000 Cells, and reported reactivity for Human and Mouse samples. Storage is at 4°C for 6 Months. The kit is described as convenient, lysate-free, high throughput and sensitive, suitable for measuring relative TBX2 levels in cultured cells and for screening the effects of treatments, inhibitors such as siRNA or chemicals, or activators on TBX2.

Two normalization approaches are provided: a monoclonal antibody specific for human GAPDH serves as an internal positive control for normalizing target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density for further normalization. The product falls within the Epigenetics and Nuclear Signaling research area. TBX2 (T-box transcription factor TBX2) localises to the nucleus, functions in DNA binding, and has documented roles in cardiac muscle development, embryonic morphogenesis, heart looping, cell aging, and the regulation of cell proliferation and transcription.
