Tau (Phospho-Thr534/217)Colorimetric Cell-Based ELISA Kit

Assay Genie supplies a Cell-Based ELISA Kit for detecting and quantifying phosphorylated tau protein directly in cultured cells, without a separate lysate preparation step. The kit targets the Tau (Phospho-Thr534/217) phospho-epitope, relevant to research into neurodegenerative disorders such as Alzheimer's disease, Pick's disease, frontotemporal dementia, cortico-basal degeneration and progressive supranuclear palsy. The associated gene is MAPT, with UniProt accession P10636.

The assay uses a colorimetric detection method read at 450 nm and is reactive with human, mouse and rat samples. It is intended for adherent cells and reports a dynamic range of more than 5000 cells. The kit is configured for 96-well microplate workflows and works on a qualitative indirect ELISA principle: phospho-Tau-specific primary antibodies are captured, and HRP-conjugated secondary antibodies bound to the Fc region of the primary antibody catalyse a colour reaction upon substrate addition.

Two normalization approaches are supported. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance, and a Crystal Violet whole-cell staining step can be used to determine cell density so that absorbance values can be corrected for plating differences.
