Description

A Colorimetric Cell-Based ELISA Kit from Assay Genie is designed for the qualitative detection of phosphorylated tau at threonine 231 in cell samples. The kit targets Tau (Phospho-Thr231) and is reactive with human, mouse, and rat samples, with a stated dynamic range of > 5000 Cells. Detection is read colorimetrically at 450 nm via an indirect ELISA format, in which target-specific primary antibodies capture phosphorylated tau and HRP-conjugated secondary antibodies catalyze the colorimetric substrate reaction.

This lysate-free, high-throughput assay allows measurement of relative phosphorylated tau levels in cultured cells and can be used to screen the effects of treatments, inhibitors such as siRNA or chemicals, or activators on tau phosphorylation. A monoclonal anti-GAPDH antibody is included to serve as an internal positive control for normalizing target absorbance, while the Crystal Violet staining method can be used to determine cell density and adjust for plating differences after readout. The kit is suitable for adherent cell samples and is provided in a 96-well microplate format.

The target, microtubule-associated protein tau (MAPT; UniProt ID P10636; Gene ID 4137), is a cytoskeletal protein that regulates microtubule assembly and stability. MAPT mutations are associated with neurodegenerative disorders including Alzheimer's disease, Pick's disease, frontotemporal dementia, cortico-basal degeneration, and progressive supranuclear palsy.
