Tau (Phospho-Thr181)Colorimetric Cell-Based ELISA Kit

This cell-based ELISA kit from Assay Genie is designed for the qualitative detection of Tau protein phosphorylated at Threonine 181 in cultured cells. Using a colorimetric detection method read at 450 nm and reactive with human samples, the kit provides a sensitive, high-throughput, lysate-free workflow. It can be used to measure relative levels of phosphorylated Tau in cultured cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on Tau phosphorylation, with a dynamic range of > 5000 cells.

The assay uses an indirect ELISA format in which phospho-specific primary antibodies capture the target, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density. The target is the microtubule-associated protein tau (MAPT; UniProt ID: P10636; Gene ID: 4137), located on chromosome 17q21.1 and involved in the establishment and maintenance of neuronal polarity. The kit is supplied in a 96-well microplate format and is intended for use with adherent cells.
