Description

Manufactured by Assay Genie, this Cell-Based ELISA Kit provides a colorimetric method for detecting phosphorylated tau at serine 422 directly in cultured cells. The assay uses an indirect ELISA format: target protein is captured by phospho-specific and total primary antibodies, with HRP-conjugated secondary antibodies catalysing a colorimetric reaction read at 450 nm. Designed as a lysate-free, high-throughput platform, it allows measurement of relative Tau phosphorylation and expression profiles in cells and can be used to screen the effects of treatments, siRNA, chemicals, or other modulators on Tau phosphorylation.

Reactive with human, mouse, and rat samples, the kit has a dynamic range of > 5000 cells. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance, and a Crystal Violet whole-cell staining step can be used to determine cell density for additional normalization of absorbance values to plating differences.

Phosphorylated tau at Ser422 is implicated in the pathogenesis of neurodegenerative diseases such as Alzheimer's and other tauopathies, making the kit relevant to research on disease progression and potential therapeutic targets. The target protein is encoded by the MAPT gene (Gene ID 4137; UniProt ID P10636).
