Description

A colorimetric cell-based ELISA kit from Assay Genie designed for the qualitative detection of Tau protein phosphorylated at serine 404 in cultured cellular samples. The assay is lysate-free and uses an indirect ELISA format in which phospho-specific primary antibodies capture the target while HRP-conjugated secondary antibodies catalyse a colour reaction upon substrate addition, with absorbance read at 450 nm.

The kit can be used to measure relative levels of phosphorylated Tau in cultured cells and to screen the effects of treatments, siRNA, chemicals, or activators on Tau phosphorylation. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalisation, and a Crystal Violet whole-cell staining step may be used to determine cell density and adjust for plating differences. Reactivity is reported for human, mouse, and rat samples, with a stated dynamic range of > 5000 Cells. The assay is configured for adherent cell samples.

Background on the target: MAPT (Gene ID 4137; UniProt P10636), a microtubule-associated protein reported at approximately 46 kDa, in which abnormal phosphorylation at serine 404 has been linked to tau tangle formation in neurodegenerative disease, including Alzheimer's disease.
