Description

From Assay Genie, this cell-based ELISA kit enables sensitive detection of phosphorylated Tau protein at serine 262 in cell lysates. The colorimetric assay is designed to support research into neurodegenerative diseases such as Alzheimer's and other tauopathies, where abnormal Tau phosphorylation is linked to the formation of neurofibrillary tangles. The kit is reactive with human, mouse, and rat samples, has a dynamic range of > 5000 Cells, and is read at 450 nm.

The assay uses an indirect ELISA format. Phospho-specific primary antibodies capture the target, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, allowing HRP to catalyse a colour change upon substrate addition. A monoclonal anti-GAPDH antibody is included to serve as an internal positive control for normalising absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density. The protocol is designed for adherent cells, with 200 µL of 20,000 cells seeded per well; 4% formaldehyde is used to fix adherent cells and 8% formaldehyde for suspension or loosely attached cells.

Providing a lysate-free, high-throughput workflow, the kit can be used to measure relative levels of phosphorylated Tau in cultured cells and to screen the effects of treatments, siRNA, chemicals, or activators on Tau phosphorylation. The target is encoded by the MAPT gene.
