Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the precise measurement of tau protein levels in cell culture samples, providing a lysate-free, high-throughput and sensitive assay for the target protein expression profile in cells. It is reactive with Human, Mouse, and Rat samples, uses adherent cells as the sample type, and is well suited to research into neurodegenerative disorders such as Alzheimer's disease.

Detection is colorimetric, read at 450 nm, with a dynamic range of > 5000 Cells. The 96-Well Microplate format uses an indirect ELISA approach in which Tau is captured by Tau-specific primary antibodies and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density. The qualitative format allows measurement of relative Tau amounts in cultured cells and supports screening of the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on Tau.

Storage is at 4°C with a stated stability of 6 Months. The target, microtubule-associated protein tau, is encoded by the MAPT gene (UniProt P10636) and is a microtubule-associated protein that regulates microtubule assembly and stability, with mutations implicated in Alzheimer's disease, Pick's disease, frontotemporal dementia, cortico-basal degeneration and progressive supranuclear palsy.
