Description

Assay Genie provides a cell-based ELISA for the qualitative detection of synuclein protein expression in cultured cells. The assay uses a lysate-free, indirect ELISA format in which synuclein is captured by target-specific primary antibodies and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. Substrate addition produces a colorimetric reaction, allowing researchers to measure relative synuclein levels and to screen the effects of treatments, inhibitors, or activators on synuclein expression.

The kit is supplied in a 96-well microplate format with colorimetric detection at 450 nm and is intended for use with adherent cells. Reactivity has been validated for Human, Mouse, and Rat samples. The dynamic range supports work with more than 5000 cells, and a monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance. For cell-density normalisation, a Crystal Violet whole-cell staining step can be performed after the colorimetric read-out, allowing absorbance values to be adjusted for plating differences. Included reagents cover primary and HRP-conjugated secondary antibodies, wash, blocking, and quenching buffers, SDS solution, ready-to-use substrate, stop solution, and adhesive plate seals. Storage stability is 4°C for 6 months.

Because synuclein is a key protein implicated in neurodegenerative disorders such as Parkinson's disease, this cell-based ELISA is well suited to studying disease mechanisms, biomarker behaviour, and potential therapeutic strategies. Its qualitative read-out is designed for comparative work—for example, comparing synuclein expression between treated and untreated cultures—rather than for absolute quantification.
