Description

This cell-based ELISA kit from Assay Genie is designed for the accurate detection of phosphorylated alpha-synuclein at tyrosine 136 in cell lysates and tissue homogenates. The assay uses a colorimetric detection method with absorbance measurement at 450 nm and has a dynamic range of greater than 5000 cells. It is reactive with human, mouse and rat samples and is suitable for use with adherent cells.

The kit employs a lysate-free, high-throughput indirect ELISA format. A phospho-specific primary antibody captures Synuclein-alpha (Phospho-Tyr136), and an HRP-conjugated secondary antibody catalyses a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step allows cell density to be determined so that absorbance values can be normalised to cell numbers. The kit can be used for measuring relative levels of phosphorylated Synuclein-alpha in cultured cells and for screening the effects of various treatments, inhibitors or activators on Synuclein-alpha phosphorylation.

Phosphorylation of alpha-synuclein at tyrosine 136 has been implicated in neurodegenerative disorders such as Parkinson's disease. The corresponding gene is SNCA, a member of the synuclein family that is abundantly expressed in the brain and linked to the pathogenesis of Parkinson's disease as well as being a major component of amyloid plaques in Alzheimer's disease.
