Synuclein-alpha (Phospho-Ser129)Colorimetric Cell-Based ELISA Kit

This cell-based ELISA kit from Assay Genie is designed for the detection of phosphorylated Synuclein-alpha at Ser129 in cultured adherent cells. Using an indirect ELISA format, the assay provides qualitative determination of Synuclein-alpha (Phospho-Ser129) levels by capturing the target with phospho-specific primary antibodies and employing HRP-conjugated secondary antibodies to generate a colorimetric signal measured at 450 nm.

The kit is reactive with human, mouse and rat samples. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step allows absorbance values to be normalized to cell density, correcting for plating differences. The assay is lysate-free and high throughput, and is suitable both for measuring the relative amounts of phosphorylated Synuclein-alpha in cultured cells and for screening the effects of various treatments, inhibitors such as siRNA or chemicals, and activators on Synuclein-alpha phosphorylation.

The dynamic range is greater than 5000 cells. Synuclein-alpha is encoded by the SNCA gene (Gene ID: 6622, UniProt ID: P37840). Phosphorylated alpha-synuclein is a key protein implicated in the pathogenesis of neurodegenerative disorders such as Parkinson's disease, and SNCA is also associated with Lewy body dementia.
