Description

A cell-based immunoassay from Assay Genie designed for the qualitative detection of synaptophysin protein in cultured cells. The kit uses an indirect ELISA format in which synaptophysin is captured by target-specific primary antibodies, and the bound primary antibody is in turn detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction readable at 450 nm.

It is described as a lysate-free, high-throughput and sensitive assay suited to measuring the relative amounts of synaptophysin in cultured cells and to screening the effects that various treatments, inhibitors (such as siRNA or chemicals) or activators have on synaptophysin expression. The kit has been validated for reactivity with human, mouse and rat samples, with a stated dynamic range of > 5000 cells, and is stored at 4 °C for 6 months. A monoclonal anti-GAPDH antibody is included as an internal positive control to normalise target absorbance, and a Crystal Violet whole-cell staining step can be used to determine cell density for further normalisation against plating differences.

Synaptophysin is an integral membrane glycoprotein of small synaptic vesicles in brain and endocrine cells, involved in synaptic vesicle trafficking and neurotransmitter release. The Assay Genie kit positions it as a biomarker of interest in research into neurological conditions such as Alzheimer's disease and Parkinson's disease, in which synaptophysin dysregulation has been reported.
