Description

From Assay Genie, this Cell-Based ELISA kit is designed for the accurate detection of phosphorylated Ser62 levels of Synapsin1 in cell lysates and tissue lysates. It offers a convenient, lysate-free, high throughput and sensitive workflow that can detect Synapsin1 protein phosphorylation and the expression profile in cells, with a stated dynamic range of > 5000 Cells. The kit is suitable for measuring the relative amounts of phosphorylated Synapsin1 in cultured cells, and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators on Synapsin1 phosphorylation. It is configured for use with adherent cells.

Reactivity covers Human, Mouse and Rat samples. Detection is colorimetric, with absorbance read at 450 nm. Qualitative determination is achieved through an indirect ELISA format: Synapsin1 (Phospho-Ser62) is captured by specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and the Crystal Violet whole-cell staining method can be used to determine cell density after the colorimetric measurement, allowing absorbance values to be normalised to cell amounts.

Synapsin1 is a neuronal phosphoprotein that associates with the cytoplasmic surface of synaptic vesicles and binds to the cytoskeleton, playing a role in the regulation of neurotransmitter release and in axonogenesis and synaptogenesis. The target is encoded by the SYN1 gene (Gene ID: 6853; UniProt ID: P17600), with a molecular weight of 70,033 Da. Phosphorylation of Ser62 on Synapsin1 has been linked to synaptic activity and regulation, making it a relevant biomarker for studying synaptic function in neurological conditions including Alzheimer's disease, Parkinson's disease and schizophrenia.
