SYK (Phospho-Tyr525)Fluorometric Cell-Based ELISA Kit

Designed by Assay Genie for phospho-protein analysis, this Fluorometric Cell-Based ELISA Kit is a convenient, lysate-free, high-throughput and sensitive assay that monitors SYK protein phosphorylation and expression profile in cells. The kit measures the relative amounts of phosphorylated SYK in cultured cells and supports screening for the effects of various treatments, inhibitors (such as siRNA or chemicals) and activators on SYK phosphorylation.

Detection is performed in an indirect ELISA format: SYK (Phospho-Tyr525) is captured by target-specific primary antibodies, and Dye 1- and Dye 2-conjugated secondary antibodies bind the Fc region of the primary antibodies to produce a fluorometric read-out upon excitation. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target RFU values, and an antibody against the non-phosphorylated counterpart of SYK (Phospho-Tyr525) is supplied for additional normalisation.

The assay is reactive with human, mouse and rat samples and uses cells as the sample type, with a dynamic range of > 5000 cells. Storage is at 4°C with 6 months of stability. SYK is a cytoplasmic tyrosine kinase of the SYK family containing two SH2 domains; it plays a central role in the B cell receptor (BCR) response and acts as an upstream activator of the PI3K, PLCgamma2 and Rac/cdc42 pathways.
