Description

From Assay Genie, this colorimetric cell-based ELISA kit is designed for the accurate and reliable detection of phospho-SYK levels in cell lysates and tissue samples. The assay targets SYK phosphorylated at Tyr525, a critical regulatory site involved in immune cell activation and signalling pathways. The kit is reactive with Human, Mouse, and Rat samples and is suitable for adherent cells, operating with a dynamic range of > 5000 Cells using a colorimetric readout at 450 nm.

The kit uses a lysate-free, high-throughput indirect ELISA format. Phosphorylated SYK (Tyr525) is captured by a target-specific primary antibody, and an HRP-conjugated secondary antibody binds the Fc region of the primary, catalysing a colorimetric reaction upon substrate addition. The assay is suitable for measuring relative amounts of phosphorylated SYK in cultured cells and for screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on SYK phosphorylation. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalisation, and a Crystal Violet whole-cell staining step may be used to determine cell density for further normalisation.

SYK is a cytoplasmic tyrosine kinase of the Syk family containing two SH2 domains, playing a central role in the B cell receptor (BCR) response and acting as an upstream activator of the PI3K, PLCgamma2, and Rac/cdc42 pathways. Dysregulation of SYK signalling has been implicated in autoimmune diseases, allergies, and cancer, making it a valuable target for therapeutic intervention. The associated gene entry is Gene ID 6850 (UniProt ID P43405).
