Description

From Assay Genie, this cell-based ELISA kit is designed for the reliable detection of phosphorylated SYK protein at tyrosine 348 in adherent cell samples. The kit uses a colorimetric detection method at 450 nm in an indirect ELISA format, in which target-specific primary antibodies are bound by HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. Reactivity covers Human, Mouse, and Rat samples, with a dynamic range of > 5000 Cells.

Designed as a lysate-free, high-throughput and sensitive assay, the kit supports measurement of relative phosphorylated SYK levels in cultured cells and screening for the effects of various treatments, inhibitors or activators on SYK phosphorylation. A monoclonal anti-GAPDH antibody is included to serve as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density, with results analysed by normalizing absorbance values to cell amounts.

SYK is a cytoplasmic tyrosine kinase of the SYK family containing two SH2 domains. It plays a central role in the B cell receptor response and acts as an upstream activator of the PI3K, PLCgamma2 and Rac/cdc42 pathways. Phosphorylation at tyrosine 348 regulates SYK activity and downstream signalling, and aberrant SYK phosphorylation has been implicated in cancer, autoimmune disorders and inflammatory conditions. The human gene maps to chromosomal location 9q22 (Gene ID: 6850; UniProt ID: P43405).
