Description

From Assay Genie, this cell-based ELISA kit enables sensitive detection of SYK (spleen tyrosine kinase) protein expression in cultured cells, supporting the measurement of relative SYK levels and the screening of treatments, inhibitors, or activators on SYK. Intended for use with adherent cells, the assay operates with a dynamic range of > 5000 Cells and uses a Colorimetric 450 nm detection method. Storage is specified at 4°C for 6 Months.

The qualitative assay is based on an indirect ELISA format: SYK is captured by SYK-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. For data normalisation, two approaches are provided. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalising the target absorbance values. Following the colorimetric measurement of HRP activity, a Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalised to cell amounts and correcting for plating differences.

SYK is a cytoplasmic non-receptor tyrosine kinase of the SYK family containing two SH2 domains, playing a central role in the B cell receptor (BCR) response and acting as an upstream activator of the PI3K, PLCgamma2, and Rac/cdc42 pathways. It is required for the sequential events of Fc gamma IIa receptor-mediated phagocytosis and is widely expressed in hematopoietic cells, where it couples activated immunoreceptors to downstream signalling events that mediate diverse cellular responses including proliferation, differentiation, and phagocytosis. The kit is reactive with human samples, and two splice variant isoforms of SYK have been described.
