Description

A phospho-specific, cell-based immunoassay from Assay Genie enables qualitative detection of Survivin phosphorylation at threonine 34 in cultured cells. Survivin is a member of the inhibitor of apoptosis (IAP) gene family, encoded by BIRC5, that is expressed during the G2/M phase of the cell cycle and is involved in cell division, apoptosis regulation, and cancer progression.

The assay uses a lysate-free, indirect ELISA format in which a phospho-specific primary antibody captures Survivin (Phospho-Thr34), and an HRP-conjugated secondary antibody catalyses the colorimetric reaction measured at 450 nm. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and Crystal Violet whole-cell staining can be used to determine cell density after the colorimetric measurement. The kit is reactive with Human, Mouse, and Rat samples and has a dynamic range of > 5000 Cells.

Classified under the Cell Death research area with a Phospho sub-type, the kit is supplied in a 96-well microplate format suited to high-throughput screening of treatments, inhibitors, or activators that affect Survivin phosphorylation.
