Description

From Assay Genie, this cell-based ELISA kit enables detection and quantification of SURF1 protein expression in cell lysates using a colorimetric method with a 96-well microplate format. It is reactive with human, mouse, and rat samples, has a dynamic range of > 5000 cells, and is stored at 4°C for up to 6 months.

The kit employs an indirect ELISA format in which SURF1-specific primary antibodies capture the target protein and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values. Crystal Violet whole-cell staining can be used to determine cell density, allowing absorbance values to be normalized to cell amounts to adjust for plating differences.

SURF1 is a protein localized to the inner mitochondrial membrane that is involved in the biogenesis of the cytochrome c oxidase (COX) complex, an essential component of the mitochondrial respiratory chain. Defects in the SURF1 gene have been linked to Leigh syndrome, a severe neurological disorder characterized by bilaterally symmetrical necrotic lesions in subcortical brain regions and commonly associated with systemic cytochrome c oxidase deficiency. The kit offers a lysate-free, high-throughput approach for analysing SURF1 expression in adherent cells, providing insights into mitochondrial function and disease pathology.
