Description

From Assay Genie, this Cell-Based ELISA kit enables qualitative detection of STEA3 protein expression in cultured adherent cells through a colorimetric method measured at 450 nm. The kit is reactive with Human, Mouse, and Rat samples and is described as lysate-free, high-throughput, and sensitive, with a stated dynamic range of > 5000 Cells. It is supplied in a 96-Well Microplate format rated for storage at 4°C for 6 Months.

The assay uses an indirect ELISA format in which STEA3-specific primary antibodies capture the target and HRP-conjugated secondary antibodies bind the Fc region of those primaries, catalysing a colorimetric reaction upon substrate addition. To support normalisation, a monoclonal antibody specific for human GAPDH is included as an internal positive control, and the Crystal Violet whole-cell staining method may be used to determine cell density after the colorimetric read. The kit is grouped under the Cell Death research area.

STEAP3 (metalloreductase STEAP3) is a multi-pass membrane protein induced directly by p53. It localises to vesicular-like structures at the plasma membrane and around the nucleus and is highly expressed in hematopoietic tissues. Functionally, it serves as an endosomal ferrireductase that reduces Fe(3+) to Fe(2+) to support transferrin-dependent iron uptake in erythroid cells, and can also reduce Cu(2+) to Cu(1+), indicating a role in copper homeostasis. The protein participates in biological processes including apoptosis, protein secretion, transferrin transport, copper ion import, and the cell cycle, and it facilitates exosome secretion in the DNA damage-induced p53-dependent pathway.
