The STAT5A Transcription Factor Activity Assay from Assay Genie is a DNA-binding ELISA used to quantify functional STAT5A binding activity in nuclear and cell lysates. STAT5A is phosphorylated by JAK kinases downstream of cytokine, growth hormone, and prolactin receptors, then dimerizes and translocates into the nucleus to drive transcription of target genes involved in immune regulation and hematopoiesis.

Wells of the microplate are pre-coated with a double-stranded oligonucleotide bearing the STAT5A consensus binding site, such as a GAS (gamma-interferon-activated sequence) motif. Activated STAT5A from added nuclear extracts binds to the immobilized probe and is detected with a STAT5A-specific antibody, yielding a chromogenic readout through TMB substrate. A typical reaction uses approximately 20 uL of nuclear extract per well, allowing direct comparison of STAT5A activity between stimulated and unstimulated samples.

Applications include JAK-STAT pathway research, cytokine signaling studies, hematopoietic and immune cell differentiation, mammary gland biology, and oncology where aberrant STAT5A drives leukemia, lymphoma, and other proliferative disorders. The assay covers human, mouse, and rat reactivity, making it broadly useful across cell line panels, primary cell models, and in vitro stimulation experiments. Reagents are stored at -20 degrees C and equilibrated to room temperature before the assay procedure; each kit supports 96 Assays in a microplate format.
