STAT4 (Phospho-Tyr693)Fluorometric Cell-Based ELISA Kit

A fluorometric cell-based ELISA kit from Assay Genie designed for the qualitative detection of phosphorylated STAT4 at tyrosine 693 directly in cultured cells. The assay uses a lysate-free, indirect ELISA format in which STAT4 (Phospho-Tyr693)-specific primary antibodies capture the target, and Dye 1- and Dye 2-conjugated secondary antibodies bound to the Fc region of the primary antibody emit light at the appropriate wavelengths to enable fluorometric detection.

Normalisation of the resulting RFU values is supported by two reagents included in the kit: a monoclonal antibody specific for human GAPDH, which serves as an internal positive control, and an antibody against the non-phosphorylated counterpart of STAT4, allowing phospho-STAT4 readings to be normalised against total STAT4. The kit is reactive with Human, Mouse and Rat samples, with a stated dynamic range of > 5000 cells. It is intended for monitoring STAT4 phosphorylation and expression profiles in cells, and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals) and activators on STAT4 phosphorylation.

STAT4 is a transcription factor of the STAT family that is phosphorylated and activated by receptor-associated kinases downstream of cytokines and growth factors, and is essential for mediating responses to IL12 in lymphocytes and for regulating the differentiation of T helper cells. The target has a chromosomal location of 2q32.2-q32.3 and a molecular weight of 85,941 Da, and is referenced under Gene ID 6775, UniProt ID Q14765 and NCBI Gene Symbol STAT4.
