STAT4 (Phospho-Tyr693)Colorimetric Cell-Based ELISA Kit

This cell-based ELISA kit from Assay Genie provides qualitative detection of phosphorylated STAT4 at tyrosine 693 in cultured cells. Using a lysate-free, indirect ELISA format, the assay captures STAT4 (Phospho-Tyr693) with target-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody and catalyze a colorimetric reaction upon substrate addition, read at 450 nm. The assay has a dynamic range of > 5000 cells and is reactive with Human, Mouse, and Rat samples.

The kit is suited for measuring the relative amounts of phosphorylated STAT4 in cultured cells and for screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on STAT4 phosphorylation. It is intended for adherent cells. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and the Crystal Violet whole-cell staining method may be used to determine cell density and adjust for plating differences.

STAT4 is a transcription factor of the STAT family, phosphorylated and activated by receptor-associated kinases downstream of cytokines and growth-factor receptors. It is essential for mediating responses to IL12 in lymphocytes and regulating the differentiation of T helper cells, with dysregulation implicated in autoimmune diseases such as rheumatoid arthritis and multiple sclerosis.
