STAT4 Colorimetric Cell-Based ELISA Kit

Assay Genie's cell-based ELISA kit is designed for the precise and reliable detection of STAT4 levels in cell lysates and tissue extracts. STAT4 is a key transcription factor that plays a critical role in the regulation of immune responses and inflammatory processes, and its dysregulation has been linked to autoimmune diseases such as rheumatoid arthritis and systemic lupus erythematosus. The kit uses an indirect ELISA format in which STAT4 is captured by target-specific primary antibodies; HRP-conjugated secondary antibodies then bind the Fc region of the primary antibody and catalyse a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control, and Crystal Violet whole-cell staining may be used to determine cell density for normalisation.

The assay is suitable for measuring relative STAT4 amounts in cultured cells and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators. It is reactive with Human, Mouse and Rat samples, with a quoted dynamic range of > 5000 Cells and detection carried out colorimetrically at 450 nm. Recommended storage is 4°C for 6 months.

In response to cytokines and growth factors, STAT family members are phosphorylated by receptor-associated kinases and form homo- or heterodimers that translocate to the cell nucleus, where they act as transcription activators. The protein is essential for mediating responses to IL12 in lymphocytes and for regulating the differentiation of T helper cells.
