STAT3 Colorimetric Cell-Based ELISA Kit

A cell-based ELISA kit from Assay Genie designed for the detection and quantification of STAT3 activation in cell lysates and tissue extracts. The assay uses a colorimetric detection method at 450 nm in a 96-well microplate format and shows reactivity with human, mouse and rat samples. With a dynamic range of > 5000 cells, the kit provides a lysate-free, high-throughput and sensitive protocol for measuring relative STAT3 protein expression in cultured cells.

The kit employs an indirect ELISA format, in which STAT3 is captured by target-specific primary antibodies and detected via HRP-conjugated secondary antibodies. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density and adjust for plating differences. The kit is suitable for screening the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators on STAT3 expression, supporting both basic and translational research into cellular signalling pathways relevant to cancer, autoimmune and inflammatory conditions.

STAT3 is a transcription factor of the STAT family that is phosphorylated and activated by receptor-associated kinases downstream of cytokines and growth factors, and is constitutively active in a number of human tumours. The kit is intended for use with adherent cells and is stored at 4°C with a stated stability of 6 months.
