The STAT1 (Phospho-Tyr701) Transcription Factor Activity Assay from Assay Genie is a DNA-binding immunoassay that quantifies the DNA-binding capacity of phosphorylated STAT1 in nuclear and cell lysates. STAT1 is a central mediator of JAK-STAT signal transduction activated by tyrosine phosphorylation at residue 701 following interferon-alpha/beta or interferon-gamma stimulation. The microplate is pre-coated with a double-stranded oligonucleotide containing the STAT consensus GAS element (TTCCGGAA). Activated STAT1 from nuclear extracts binds this consensus sequence and is detected with a phospho-Tyr701 specific antibody, followed by HRP substrate and colorimetric readout. This Assay Genie kit supports research into cytokine signaling, antiviral immunity, and inflammatory pathways in human, mouse, and rat samples. Recommended workflow uses 10 uL of nuclear extract per well; store all reagents at -20 degrees C and aliquot to limit freeze-thaw cycles. The assay is for research use only and provides a quantitative measure of transcription factor activation independent of total STAT1 protein levels.
