Description

A cell-based ELISA kit from Assay Genie is designed for the accurate detection and quantification of STAT1 phosphorylated at tyrosine 701 in cell lysates. Phosphorylation of STAT1 at Tyr701 is required for its activation and subsequent regulation of gene expression, and the kit supports research into diseases where this pathway is dysregulated, including cancer, autoimmune disorders, and infectious diseases.

The assay uses a colorimetric detection method at 450 nm in an indirect ELISA format, with a stated dynamic range of > 5000 Cells. Reactivity is reported for human, mouse, and rat. The lysate-free, high-throughput protocol enables measurement of relative STAT1 phosphorylation in cultured cells, allowing researchers to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators. The sample type is adherent cells.

A monoclonal GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method is used to determine cell density. STAT1 (Phospho-Tyr701) is captured by target-specific primary antibodies, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition.
