STAT1 (Phospho-Ser727)Colorimetric Cell-Based ELISA Kit

From Assay Genie, this Cell-Based ELISA Kit is designed for the quantitative detection of phosphorylated STAT1 at serine 727 in adherent cell cultures. Using a colorimetric method, the assay is a lysate-free, high-throughput format in which phospho-specific primary antibodies capture the analyte while HRP-conjugated secondary antibodies catalyse a colourimetric reaction upon substrate addition. The kit can be used to measure relative levels of phosphorylated STAT1 in cultured cells and to screen the effects of various treatments, inhibitors (e.g., siRNA or chemicals), or activators on STAT1 phosphorylation.

Reactivity is established for Human, Mouse and Rat samples, with a dynamic range of > 5000 cells and detection read at 450 nm. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and Crystal Violet whole-cell staining may subsequently be used to determine cell density so that absorbance values can be adjusted for plating differences. STAT1 is a transcription factor of the STAT family that, upon phosphorylation, forms homo- or heterodimers that translocate into the nucleus to regulate transcription, with phosphorylation at Ser727 specifically regulating its nuclear translocation and transcriptional activity in cytokine signalling and antiviral defence.
