STAT1 Colorimetric Cell-Based ELISA Kit

From Assay Genie, this Cell-Based ELISA is a sensitive, high-throughput, lysate-free assay designed for the precise measurement of STAT1 levels in cell lysates, serum, and tissue culture supernatants. STAT1 (Signal Transducer and Activator of Transcription 1) is a key transcription factor involved in immune responses and cellular growth regulation, and its dysregulation has been linked to cancer, autoimmune disorders, and infectious diseases, making it a valuable biomarker for research into disease mechanisms and targeted therapies.

The kit uses an indirect ELISA format in which STAT1 is captured by STAT1-specific primary antibodies. HRP-conjugated secondary antibodies then bind the Fc region of the primary antibody, and the bound HRP enzyme catalyzes a colorimetric reaction upon substrate addition, with detection at 450 nm. The assay is reactive with human, mouse, and rat samples, has a dynamic range of > 5000 Cells, and is intended for storage at 4°C for 6 months.

Suitable for measuring the relative amounts of STAT1 in cultured cells, the kit can also be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on STAT1 expression. Two normalisation approaches are supported: a monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and Crystal Violet whole-cell staining can be used to determine cell density and adjust for plating differences.
