StableScript (250 U)

A reverse transcriptase from QIAGEN designed for one-step RT-qPCR and long-range RT-PCR applications. The enzyme shows no detectable RNase H activity and offers high thermostability, performing across a temperature range of 45° to 65°C. This combination helps overcome detection challenges with difficult RNA targets and supports high cDNA yields, with cDNA transcripts reported up to 12.3 kb. The enzyme also demonstrates reliable performance in the presence of common inhibitors.

One unit is defined as the amount of enzyme required to incorporate 1 nmol of dTTP into acid-insoluble material in 30 minutes at 55°C using poly r(A)/oligo(dT) as a substrate. The protein is sourced from Equine Infectious Anemia Virus. The enzyme is supplied in 20 mM KPO4, 1 mM EDTA, 1 mM DTT, stabilizer and 50% glycerol at pH 7.0 (25°C), together with a 4X StableScript Reaction Buffer.

Quality control testing confirms physical purity greater than 99%, with single-stranded exonuclease activity releasing less than 5% and double-stranded exonuclease activity releasing less than 1% in 5000 U samples. No double-stranded endonuclease conversion is observed, E. coli DNA contamination is below 10 copies per 5000 U, and no non-specific RNase is detectable. Functional performance is verified by amplification of test lots within 1 Ct of the reference lot in a one-step RT-qPCR assay. Listed applications include cDNA synthesis, one-step RT-qPCR, long-range RT-PCR and ligation-based adapter addition for RNA sequencing.
