For gene expression analysis using real-time two-step RT-PCR, QIAGEN's QuantiNova Reverse Transcription Kit delivers a fast cDNA synthesis workflow with integrated genomic DNA removal and an additional internal control. Genomic DNA contamination in RNA samples is effectively eliminated by the included gDNA Removal Mix, removing the need for a separate DNase digestion during or after RNA purification and eliminating the need to design RNA-specific primers or probes. The kit comprises the gDNA Removal Mix, Internal Control RNA, QuantiNova Reverse Transcriptase, QuantiNova RT buffer system, and RT Primer Mix.

The QuantiNova Reverse Transcriptase has a high affinity for RNA and supports cDNA synthesis from a wide range of RNA amounts (10 pg – 5 μg). The RT Primer Mix enables cDNA synthesis from all regions of transcripts, including 5' regions. Difficult templates with high GC-content or complex secondary structure are successfully reverse transcribed, as the reaction proceeds at 42°C and the enzyme is then inactivated at 85°C. Additional steps for RNA denaturation or RNase H digestion are not required, and the QuantiNova RT Buffer has been optimized to be compatible with real-time PCR buffers.

Genomic DNA removal and cDNA synthesis are completed in just 20 minutes. The internal control RNA is a defined transcript that can be optionally added to samples and transcribed into cDNA, intended to report instrument or chemistry failures, errors in assay setup, and the presence of inhibitors.
