A reverse transcription kit from QIAGEN enables fast cDNA synthesis for sensitive real-time two-step RT-PCR in gene expression analysis. The system supports a wide range of RNA template amounts, from 10 pg to 1 µg per reaction, allowing quantification of even low-abundance transcripts.

Contaminating genomic DNA is eliminated using the included gDNA Wipeout Buffer, removing the need for a separate DNase digestion during or after RNA purification. Both genomic DNA removal and cDNA synthesis are completed in 20 minutes, run at a single incubation temperature of 42°C with inactivation at 95°C. Quantiscript Reverse Transcriptase is a blend of Omniscript and Sensiscript Reverse Transcriptases, delivering high RNA affinity for efficient cDNA synthesis. Even difficult templates with high GC-content or complex secondary structure are successfully reverse transcribed. An optimized RT Primer Mix of oligo-dT and random primers enables cDNA synthesis from all regions of RNA transcripts, including 5' regions.

The kit is suitable for starting materials including laser-microdissected samples and tissue biopsies, and has been tested for reverse transcription of bacterial RNA when paired with the RNeasy Mini Kit. The Quantiscript RT Buffer is compatible with real-time PCR buffer. For optimal amplification efficiency in downstream qPCR, amplicon length should be less than 150 bp. The kit should be stored at -20°C upon receipt.
