Description

A high-throughput Cell-Based ELISA kit from Assay Genie provides sensitive, lysate-free detection of SSXT protein expression in cultured cells. Using an indirect ELISA format, SSXT is captured by target-specific primary antibodies, which are then bound by HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. The assay is read colorimetrically at 450 nm.

Reactive with human, mouse, and rat samples, the kit supports a dynamic range of > 5000 cells and is supplied in a 96-well microplate format with storage at 4°C for 6 months. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and the Crystal Violet whole-cell staining method may be used to determine cell density. Absorbance values can be normalized to cell amounts to adjust for plating differences. The kit supports measuring relative SSXT levels in cultured cells and screening the effects of various treatments, inhibitors, or activators, with applications in epigenetics and nuclear signaling research.

SSXT (also known as SS18 or SYT) is encoded by the SS18 gene at chromosomal region 18q11.2 (Gene ID: 6760; UniProt: Q15532; OMIM: 600192). The reported molecular weight is 418. The protein functions as a transcriptional coactivator, acting synergistically with RBM14 in nuclear receptor coactivation and general transcriptional coactivation, and is classified as an oncoprotein. A chromosomal aberration involving SS18 is implicated as a cause of synovial sarcoma, with the SSXT-SSX1 or SSXT-SSX2 fusion products considered responsible for transforming activity.
