Description

From Assay Genie, this colourimetric cell-based ELISA kit is designed for the qualitative detection of SSB (Lupus La protein) in cultured human cells using an indirect ELISA format. The lysate-free assay captures SSB with target-specific primary antibodies, and HRP-conjugated secondary antibodies then catalyse a colourimetric reaction upon substrate addition, with absorbance read at 450 nm. The kit can be used to measure relative SSB levels in adherent cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on SSB.

A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step allows cell density to be determined so that results can be normalised to cell amount. The format is a 96-well microplate with a stated dynamic range of > 5000 cells, and the kit is stored at 4°C for 6 months.

SSB (also known as Sjogren syndrome antigen B) is a nuclear protein involved in diverse aspects of RNA metabolism, including binding the 3' termini of nascent RNA polymerase III transcripts, processing pre-tRNA ends, and acting as an RNA chaperone. Autoantibodies reacting with this protein are found in the sera of patients with Sjogren syndrome and systemic lupus erythematosus.
