The SRY protein (Sex-determining Region Y) is the master transcriptional regulator that initiates testis development from the bipotential gonad. This DNA-binding ELISA from Assay Genie measures the functional binding activity of SRY in nuclear and cell lysates, distinguishing the active transcription factor from total protein abundance. Applications include male sex determination, gonadal differentiation, and disorders of sex development (DSD) research. The assay uses an immobilized consensus oligonucleotide probe to capture active SRY from human, mouse, or rat nuclear extracts, followed by a target-specific antibody and HRP/TMB readout in a microplate format. Reagents are shipped on dry ice and stored at -20 °C; thaw the kit components on ice and avoid repeated freeze-thaw cycles. Researchers commonly apply this assay to compare SRY activity across developmental stages, gene knockout or mutant cell models, and patient-derived samples linked to Swyer syndrome or 46,XY gonadal dysgenesis.
