Description

Designed for the quantitative detection of SRY (sex-determining region Y) protein in cell lysates and tissue homogenates, this Assay Genie Cell-Based ELISA kit uses an indirect ELISA format with HRP-conjugated secondary antibodies and colorimetric detection at 450 nm. It is reactive with human samples and intended for use with adherent cells, making it suitable for researchers studying sex determination and gonadal development.

The assay is qualitative in nature, with two normalisation methods provided. A monoclonal anti-human GAPDH antibody serves as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density and adjust for plating differences. The kit is described as convenient, lysate-free, high-throughput and sensitive, and can be used to measure relative SRY levels in cultured cells or to screen the effects of treatments, inhibitors or activators on SRY expression. SRY is a transcriptional regulator that controls the genetic switch in male development, directing the differentiation of supporting cell precursors towards Sertoli rather than granulosa cells, and defects in SRY are associated with 46,XY sex reversal type 1, 46,XX sex reversal type 1 and Turner syndrome.

Intended for use in developmental biology research, the assay has a stated dynamic range of > 5000 cells, with recommended storage at 4°C and a stated shelf life of 6 months.
