SRF (Phospho-Ser99)Colorimetric Cell-Based ELISA Kit

From Assay Genie, this colorimetric cell-based ELISA kit is designed to measure relative levels of SRF phosphorylated at Ser99 in cultured cells. The assay is reactive with human, mouse, and rat samples and supports research into serum response factor (SRF) phosphorylation, a key event in the regulation of gene expression and cell proliferation whose dysregulation has been implicated in cancer, heart disease, and neurological disorders.

Using an indirect ELISA format, the kit captures SRF (Phospho-Ser99) with a target-specific primary antibody, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance, and a Crystal Violet whole-cell staining step may be used to determine cell density. Detection is colorimetric at 450 nm with a dynamic range of > 5000 Cells, and the kit is intended for use with adherent cells in a microplate format.

SRF is a MADS-box transcription factor that binds the serum response element (SRE) and regulates transcription of immediate early genes including c-fos. The assay is suited to screening the effects of various treatments, inhibitors such as siRNA or chemicals, and activators on SRF phosphorylation, as well as exploring MAPK-linked signalling that acts through ternary complex factors.
