Description

From Assay Genie, this cell-based ELISA kit provides a lysate-free, colorimetric assay for the qualitative detection of serum response factor (SRF) directly in adherent cells. Run in a 96-well microplate format with readout at 450 nm, the kit supports high-throughput measurement of relative SRF levels in cultured cells and can be used to screen the effects of treatments, siRNA, chemical inhibitors, or activators on SRF expression. It is reactive with human, mouse, and rat samples and has a stated dynamic range of > 5000 cells, with storage stability of 4 °C for 6 months.

Detection follows an indirect ELISA format: SRF in fixed cells is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance, and a Crystal Violet whole-cell staining step is provided to determine cell density so that absorbance values can be adjusted for plating differences between wells.

SRF is a MADS-domain transcription factor that binds the serum response element (SRE) to regulate immediate-early genes such as c-fos, and is associated with cellular processes including growth, differentiation, and migration. The protein is catalogued under Gene ID 6722 and UniProt ID P11831, and according to the manufacturer, dysregulation of SRF has been implicated in cancer, cardiovascular disease, and neurodegenerative disorders, making the kit a relevant tool for research into SRF signalling pathways and potential therapeutic targets.
