Description

A Colorimetric Cell-Based ELISA Kit from Assay Genie is designed to detect phosphorylated Src at Tyrosine 216 in cell lysates and tissue homogenates. The kit is reactive with Human, Mouse, and Rat samples, with a dynamic range of > 5000 Cells. It is intended for adherent cells and uses an indirect ELISA format in which Src (Phospho-Tyr216) is captured by target-specific primary antibodies and detected using HRP-conjugated secondary antibodies, producing a colorimetric readout at 450 nm. The procedure is lysate-free and high-throughput, allowing measurement of relative phosphorylated Src levels in cultured cells and assessment of how treatments, inhibitors, or activators affect Src phosphorylation.

To support quantitative interpretation, the kit includes a monoclonal anti-GAPDH antibody for use as an internal positive control for normalization, and Crystal Violet solution for whole-cell staining to determine cell density. SRC is a proto-oncogenic cytoplasmic tyrosine kinase of the Src family, with a molecular weight of 60kDa, implicated in cell signaling related to growth, proliferation, and survival, and aberrant activation is associated with cancer, inflammatory disorders, and neurodegenerative conditions. The target corresponds to the human SRC gene (Gene ID: 6714, UniProt ID: P12931).
