Src Colorimetric Cell-Based ELISA Kit

Assay Genie offers a Cell-Based ELISA Kit designed to detect Src protein expression profile in cells using a colorimetric readout measured at 450 nm. The kit is configured in a 96-Well Microplate format, is reactive with Human, Mouse, and Rat samples, and has a stated dynamic range of > 5000 Cells. It is intended for use with adherent cells and is stored at 4°C for up to 6 months.

Detection is achieved through an indirect ELISA format in which Src is captured by Src-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method may be used after the colorimetric measurement to determine cell density and adjust for plating differences. The assay is described as lysate-free, high throughput, and qualitative, and can be applied to measuring relative Src levels in cultured cells or to screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on Src.
