SPIC Transcription Factor Activity Assay

The SPIC (Spi-C) transcription factor plays a pivotal role in immune cell lineage commitment, particularly in the differentiation of B-1 B cells, marginal zone B cells, the regulation of erythroid progenitors, and the maturation of plasmacytoid dendritic cells and tissue-resident macrophages such as splenic red pulp macrophages. This Assay Genie kit quantifies active SPIC DNA-binding capacity in nuclear extracts by immobilizing a double-stranded oligonucleotide probe containing the SPIC consensus binding sequence on the plate, allowing researchers to monitor transcriptional activation states in lysates prepared from human, mouse, and rat samples. Each kit includes pre-coated wells, a SPIC-specific detection antibody, lysis and binding buffers optimized for nuclear protein extraction, and developing reagents sufficient for 96 individual binding reactions read on a standard microplate reader. Shipped refrigerated on blue ice, the reagents should be stored at -20 °C for long-term stability, and repeated freeze-thaw cycles of the antibody components should be avoided.
