Assay Genie's SPI1 Transcription Factor Activity Assay is a DNA-binding ELISA that quantifies PU.1 (encoded by the SPI1 gene) DNA-binding activity in nuclear and cell lysates. PU.1 is an ETS-family transcription factor that recognizes the purine-rich PU-box motif (5'-GAGGAA-3') and serves as a master regulator of hematopoiesis, controlling myeloid and B-lymphocyte lineage commitment. The kit uses oligonucleotide probes immobilized on a microplate to capture active PU.1 from nuclear extracts, with detection via a specific antibody. Researchers apply this assay in studies of acute myeloid leukemia, immune cell differentiation, and transcriptional regulation across human, mouse, and rat samples. The reagents are sufficient for 96 reactions on a standard microplate format. Store reagent components at -20 degrees C; thaw on ice before use and minimize repeated freeze-thaw cycles. For in vitro research use only.
