Description

From Assay Genie, this cell-based ELISA kit is designed to measure the activity of SPI1 (PU.1), a transcription factor involved in regulating cell differentiation and immune responses, directly within cell lysates. The assay uses a colorimetric detection method read at 450 nm and is formatted for use in a 96-well microplate.

The kit employs an indirect ELISA format in which SPI1 is captured by SPI1-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. HRP activity is then measured colorimetrically upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalization, and a Crystal Violet whole-cell staining method may be used to determine cell density and adjust for plating differences. The kit is reactive with human, mouse, and rat samples, has a dynamic range of > 5000 cells, and is intended for adherent cell samples. Storage is at 4°C with a stability of 6 months.

Applications include measuring relative amounts of SPI1 in cultured cells and screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on SPI1, supporting research in immunology, oncology, epigenetics, and nuclear signaling.
